I de-tune the string then loosen the saddle screw just a bit so that there's still friction. After that it's just a matter of sliding the saddle a bit, tightening it down, tuning up, and seeing if I guessed it right. This sounds very tedious but it's really not that bad, especially since I know that I'll be good for a long while without having to touch it again.
