Do i need to do anything setup wise? Should i tighten the truss rod? Do the nut slots need to be widened?

You should not have to widen the nut slots, and only adjust the truss rod if it starts buzzing. You may have to adjust the saddles a bit because the 10's are larger and will sit higher on the saddles thereby raising the action.

^^^ What he said. More than likely, you won't even notice the difference, other than sore fingers.

I had used 10's till i got my silo, and it was time for a string change...wow, it really is tougher again. :D
Now, on one of these truss rods, i turn clockwise to tighten it, right?

and now i have more buzzing since tightening it...

That makes sense. You are flattening the fretboard when you tighten it. By now, you may have already loosened it to have some relief in the neck.
