Hi, I usually play 2221 .010-.046 and I would like to try the power slinky 2220 .011-.048. I have a Les Paul. Would I have to change the nut to accommodate the larger strings?

 
I would deepen the slots for sure. Gibson nuts are typically very high and you want relatively low string height over the 1st fret for best playability with heavier strings.
